| Product Name | Green Nucleic Acid Gel Stain (20× in Water) |
| Catalog No. | IVDRM-0110 |
| Description | Green Nucleic Acid Gel Stain (20× in Water) from Alta DiagnoTech is a DNA-binding dye with spectral properties similar to SYBR Green I. Its unique working characteristics make it suitable not only for quantitative real-time PCR (qPCR) but also for high-resolution melting (HRM) analysis. Compared with SYBR Green I, it offers three key advantages: (1) far lower PCR inhibition—an intelligent 'release-on-demand' DNA-binding technology allows higher dye concentrations for greatly enhanced amplification signals and eliminates dye redistribution, enabling both multiplex PCR and HRM analysis; (2) exceptional stability—the dye is not degraded during normal storage, handling, or PCR cycling, can be stored at room temperature or refrigerated in buffer, and tolerates repeated freeze-thaw cycles, whereas SYBR Green I is unstable and becomes more PCR-inhibitory after degradation; (3) improved safety—cell membrane penetration assays show the dye barely crosses cell membranes and is neither mutagenic nor cytotoxic. The dye is also highly compatible with all major brands of qPCR instruments, so replacing SYBR Green I requires no change to existing protocols, procedures, or equipment. |
| Intended Use | Fluorescent detection of DNA in quantitative real-time PCR (qPCR) and high-resolution melting (HRM) analysis; also suitable for multiplex PCR. |
| Principle / Technology | A DNA-binding dye with spectral characteristics similar to SYBR Green I, based on an intelligent 'release-on-demand' DNA-binding technology that minimizes PCR inhibition even at high dye concentrations. |
| Detection Method | Spectral properties similar to SYBR Green I; detected using standard SYBR Green I/FAM channels on qPCR instruments from all major brands. |
| Sample Type | DNA (double-stranded DNA binding dye). |
| Performance Range / Specifications | Far lower PCR inhibition than SYBR Green I; high dye concentrations can be used to obtain greatly enhanced amplification signals and to eliminate dye redistribution, supporting multiplex PCR and HRM analysis. Extremely stable during normal storage, handling, and PCR; tolerates repeated freeze-thaw cycles. |
| Sensitivity / LOD | High dye concentrations can be used without significant PCR inhibition, delivering greatly enhanced amplification signals compared with SYBR Green I. |
| Specificity | DNA-binding dye with spectral properties similar to SYBR Green I. |
| Reaction Conditions / Protocol | Supplied as a 20× concentrate in water. Directly replaces SYBR Green I in qPCR workflows—no changes to current operating procedures or instrumentation are required. The dye in buffer can be stored at room temperature or refrigerated and withstands repeated freeze-thaw cycles. |
| Components / Formulation | 20× concentrated DNA-binding dye solution in water. |
| Storage Conditions | Store at 4°C, dry and protected from light. The dye in buffer can be safely stored at room temperature or refrigerated and tolerates repeated freeze-thaw cycles. |
| Shelf Life | 12 months when stored as recommended. |
| Package Specifications | 500 μL |
| Product Form | Liquid; 20× aqueous solution. |
| Quality Control | Cell membrane penetration testing confirms minimal membrane permeability; performance verified against SYBR Green I for PCR inhibition, stability, and safety. |
| Concentration | 20× |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | Shelf life of 12 months at 4°C, dry and protected from light. Extremely stable: not degraded during normal storage, handling, or PCR; tolerates repeated freeze-thaw cycles. |
| Regulatory / Compliance | Cell membrane penetration assays show the dye barely crosses cell membranes; it is non-mutagenic and non-cytotoxic, offering a safer alternative to SYBR Green I. |
| Compatibility | Compatible with qPCR instruments from all major brands; a direct replacement for SYBR Green I with no change to protocols or equipment. Suitable for multiplex PCR and HRM analysis. |
| Application Notes / Precautions | Much lower PCR inhibition than SYBR Green I, allowing higher dye concentrations for stronger amplification signals. Extremely stable during normal storage, handling, and PCR; in contrast, SYBR Green I is unstable and becomes more PCR-inhibitory after degradation. Minimally membrane-permeable and safer to handle: non-mutagenic and non-cytotoxic, whereas SYBR Green I may inhibit normal DNA repair mechanisms in cells and enhance mutagenesis. High dye concentrations eliminate dye redistribution, enabling multiplex PCR and HRM analysis. |
