| Product Name | Green DNA-Binding Dye for qPCR and Gel Staining (20× in Water) |
| Catalog No. | IVDRM-0112 |
| Description | Green DNA-Binding Dye for qPCR and Gel Staining (20× in Water) from Alta DiagnoTech is a new-generation DNA-binding dye for real-time quantitative PCR (qPCR). Designed around a 'release-on-demand' DNA-binding technology, the dye is a dimer in which two monomeric dye units are linked by a flexible arm: in the absence of DNA it adopts a non-fluorescent cyclic conformation, and upon DNA binding it relaxes into a random conformation and emits intense green fluorescence. Compared with other fluorescent dyes on the market such as FAM and SYBR Green I, it offers (1) extremely high sensitivity, delivering stronger PCR amplification signals than SYBR Green I at recommended concentrations; (2) minimal PCR inhibition, making it ideal for multiplex PCR and HRM analysis—fast PCR cycling protocols can be used, higher dye concentrations yield far stronger signals, and the absence of dye redistribution between amplicons enables both multiplex PCR and high-resolution melting analysis; (3) outstanding stability, remaining intact during normal storage, handling, and PCR, with buffer solutions safely stored at room temperature or refrigerated and tolerant of repeated freeze-thaw cycles; (4) excellent safety, as cell membrane penetration assays show the dye barely crosses cell membranes and is non-mutagenic and non-cytotoxic; and (5) superior compatibility, with a spectrum similar to SYBR Green I that works with qPCR instruments from all major brands without any change to existing protocols or equipment. In addition, it is currently the only dye suitable for droplet digital PCR systems, and it also performs exceptionally in microfluidic PCR, isothermal amplification, capillary electrophoresis, DNA quantification, and cell viability assays. The product is supplied as a 20× aqueous solution. |
| Intended Use | DNA-binding fluorescent dye for real-time quantitative PCR (qPCR), multiplex PCR, high-resolution melting (HRM) analysis, and droplet digital PCR; also applicable to microfluidic PCR, isothermal amplification, capillary electrophoresis, DNA quantification, and cell viability assays. |
| Principle / Technology | Based on an intelligent 'release-on-demand' DNA-binding technology: two monomeric dye molecules are linked by a flexible arm into a dimer that forms a non-fluorescent cyclic structure without DNA; in the presence of DNA, the dimer adopts a random conformation, binds DNA, and emits strong green fluorescence. |
| Detection Method | Emits strong green fluorescence upon DNA binding; spectrum similar to SYBR Green I, detected on standard SYBR Green I/FAM channels of qPCR instruments from all major brands, with no change to existing equipment or procedures. |
| Sample Type | DNA (double-stranded DNA binding dye for PCR-based detection). |
| Performance Range / Specifications | Stronger PCR amplification signals than SYBR Green I at recommended concentrations; minimal PCR inhibition supports fast PCR protocols and higher dye concentrations; no dye migration between amplicons at recommended concentrations; stable during normal storage, handling, and PCR, and tolerant of repeated freeze-thaw cycles. |
| Sensitivity / LOD | Extremely high sensitivity: provides stronger PCR amplification signals than SYBR Green I when used at recommended concentrations. |
| Specificity | Non-fluorescent in the absence of DNA; binds DNA and emits strong green fluorescence. Spectrum similar to SYBR Green I. |
| Reaction Conditions / Protocol | Supplied as a 20× concentrate in water. Directly replaces SYBR Green I in qPCR workflows with no change to operating procedures or instrumentation; compatible with fast PCR cycling protocols and higher dye concentrations. Stable in buffer at room temperature or refrigerated and tolerates repeated freeze-thaw cycles. |
| Components / Formulation | 20× concentrated DNA-binding dye solution in water. |
| Storage Conditions | Store at 4°C, protected from light. Dye in buffer can be safely stored at room temperature or refrigerated and tolerates repeated freeze-thaw cycles. |
| Shelf Life | 24 months when stored as recommended. |
| Package Specifications | 500 µL; 1 mL |
| Product Form | Liquid; 20× aqueous solution. |
| Quality Control | Cell membrane penetration assays confirm minimal membrane permeability; verified non-mutagenic and non-cytotoxic; performance benchmarked against SYBR Green I and FAM. |
| Concentration | 20× |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | Shelf life of 24 months at 4°C protected from light. Ultra-stable during normal storage, handling, and PCR; tolerates repeated freeze-thaw cycles. |
| Regulatory / Compliance | Cell membrane penetration assays show the dye barely crosses cell membranes; non-mutagenic and non-cytotoxic. |
| Compatibility | Spectrum similar to SYBR Green I; compatible with qPCR instruments from all major brands without changes to protocols or equipment. Comparable performance to EvaGreen. Suitable for multiplex PCR, HRM analysis, and droplet digital PCR systems. |
