| Product Name | Fast Restriction Enzyme Esp3I (Recognition Site: CGTCTC) |
| Catalog No. | IVDRM-0039 |
| Description | This fast restriction enzyme is produced by recombinant genetic engineering and delivers precise, complete DNA cleavage within 5-15 minutes. It is ideal for rapid digestion of plasmid DNA, PCR products, and genomic DNA. All enzymes in the series share a single universal digestion buffer, which greatly simplifies reaction setup, and exhibit generous activity excess, allowing effortless digestion of excess substrate or difficult templates. |
| Intended Use | For rapid digestion of plasmid DNA, PCR products, and genomic DNA; well suited for Golden Gate assembly and CRISPR vector construction. |
| Principle / Technology | Recombinant fast restriction endonuclease produced by genetic engineering; recognizes a specific palindromic/defined DNA sequence and cleaves both strands at defined positions within 5-15 minutes. |
| Sample Type | Plasmid DNA, PCR products, genomic DNA, and other double-stranded DNA substrates. |
| Performance Range / Specifications | Completes precise DNA digestion within 5-15 min. Good activity excess; easily handles digestion of excess substrate or difficult templates. |
| Specificity | Specifically recognizes and cleaves the sequence 5'-CGTCTC(N)1↓-3' 3'-GCAGAG(N)5↑-5'; isoschizomers: BsmBI. |
| Reaction Conditions / Protocol | 1× universal fast digestion buffer; incubate at the optimal temperature of 37°C. |
| Storage Conditions | Store at -20°C. |
| Shelf Life | 2 years. |
| Package Specifications | 30 T |
| Concentration | 10 U/μL |
| Activity / Unit Definition | 10 U/μL |
| Source / Origin | Recombinant enzyme produced by genetic engineering. |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | 2 years. |
| Compatibility | BsmBI |
| Recommended Buffer System | All fast restriction enzymes in this series share one universal digestion buffer, simplifying multi-enzyme reaction setup. |
| Application Notes / Precautions | This enzyme is temperature-sensitive; handle and store with care. For difficult templates (e.g., lenti-CRISPRV2 plasmid), increase enzyme amount and extend digestion time: in a 20 μL reaction with 1 μg plasmid, use 3-4 μL enzyme and incubate for 2-3 h. An optional color digestion buffer containing red and yellow tracking dyes allows direct gel loading without additional loading buffer. |
| Recognition Site | 5'-CGTCTC(N)1↓-3' 3'-GCAGAG(N)5↑-5' |
| Heat Inactivation | Incubate at 80°C for 20 min. |
| Methylation Sensitivity | CpG methylation: sequence fully overlaps; cleavage is blocked. EcoBI methylation: sequence may overlap; cleavage may be affected. |
For research use only, not for clinical use.
