| Product Name | Fast Restriction Enzyme DpnI (Recognition Site: 5'-GAm6↓TC-3') |
| Catalog No. | IVDRM-0032 |
| Description | Fast Restriction Enzyme DpnI is a methylation-dependent fast restriction endonuclease produced by recombinant genetic engineering. It precisely completes DNA cleavage within 5–15 minutes and is suitable for rapid digestion of plasmid DNA, PCR products, genomic DNA, and more. All fast restriction enzymes in this series share a single universal digestion buffer, which greatly simplifies the reaction setup. In addition, the enzyme provides ample activity redundancy, allowing effortless digestion of excess substrate or difficult templates. DpnI recognizes the sequence 5'-GAm6↓TC-3' and cleaves only when the recognition site carries N6-methyladenine (dam) methylation. |
| Intended Use | Rapid digestion of plasmid DNA, PCR products, and genomic DNA in 5–15 minutes for molecular cloning and other molecular biology workflows. |
| Principle / Technology | Recombinant fast restriction endonuclease that recognizes a specific double-stranded DNA sequence and cleaves it within 5–15 minutes; a single universal fast digestion buffer is shared across the series to simplify reaction setup, with generous activity redundancy to handle excess substrate or difficult templates. |
| Sample Type | Plasmid DNA, PCR products, genomic DNA. |
| Performance Range / Specifications | Recognition site: 5'-GAm6↓TC-3' (3'-CT↑Am6G-5') Enzyme activity: 20 U/μL Heat inactivation: 80°C for 20 min |
| Specificity | Recognizes and cleaves only dam-methylated 5'-GAm6↓TC-3' / 3'-CT↑Am6G-5'; unmethylated DNA is not recognized or cut. |
| Reaction Conditions / Protocol | 1× universal fast digestion buffer; optimal incubation at 37°C. |
| Storage Conditions | Store at -20°C. |
| Shelf Life | 2 years when stored at -20°C. |
| Package Specifications | 50 T |
| Concentration | 20 U/μL |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | Stable for 2 years at -20°C. |
| Recommended Buffer System | A single universal fast digestion buffer is shared across the fast restriction enzyme series, simplifying reaction setup (use at 1×). |
| Application Notes / Precautions | DpnI is methylation-dependent: it recognizes and cleaves only DNA whose recognition site carries N6-methyladenine (dam) methylation. Unmodified PCR products (amplified DNA without methylation) cannot be recognized or cut. EcoBI methylation: the sequence may overlap and cleavage may be affected. |
| Recognition Site | 5'-GAm6↓TC-3' 3'-CT↑Am6G-5' |
| Heat Inactivation | Incubate at 80°C for 20 min. |
| Methylation Sensitivity | Methylation-dependent enzyme: cleaves only dam-methylated (N6-methyladenine) DNA; unmethylated DNA is not cut. EcoBI: sequence may overlap and cleavage may be affected. |
