Product Name Fast Restriction Enzyme BclI (Recognition Site: T/GATCA)
Catalog No. IVDRM-0016
Description Fast Restriction Enzyme BclI from Alta DiagnoTech is a recombinant rapid restriction endonuclease that performs precise DNA cleavage within 5-15 minutes. It is ideal for rapid digestion of plasmid DNA, PCR products and genomic DNA. All enzymes in the fast digestion series share one universal buffer to simplify reaction setup, and their excellent activity redundancy allows easy digestion of excess substrate or difficult templates.
Intended Use Rapid digestion of plasmid DNA, PCR products and genomic DNA for molecular cloning and related molecular biology workflows.
Principle / Technology Recombinant fast restriction endonuclease produced by genetic engineering; recognizes the specific double-stranded DNA sequence 5'-T/GATCA-3' and cleaves rapidly at 37°C in a universal fast digestion buffer.
Sample Type Plasmid DNA, PCR products, genomic DNA
Performance Range / Specifications Complete digestion within 5-15 min
Supplied at 20 U/μL
Good activity redundancy - readily digests excess substrate or difficult templates
No star activity observed after 3 h incubation
Specificity Specifically recognizes 5'-T/GATCA-3'; cleavage is blocked when the site fully overlaps Dam or EcoBI methylation, while Dcm, CpG and EcoKI methylation have no effect.
Reaction Conditions / Protocol 1× universal fast digestion buffer; optimal incubation at 37°C
Storage Conditions -25°C to -15°C
Shelf Life 2 years
Package Specifications 125 T
Concentration 20 U/μL
Source / Origin Recombinant (produced by genetic engineering technology)
Expiration Date / Stability Stable for 2 years when stored at -25°C to -15°C.
Compatibility Isoschizomers: BsiQI, FbaI, Ksp22I
Recommended Buffer System A single universal fast digestion buffer is shared across the entire fast restriction enzyme series, simplifying single- and multi-enzyme digestion setups.
Application Notes / Precautions No star activity is observed after 3 h incubation; prolonged digestion may induce star activity, so avoid unnecessarily extended incubation. Dam or EcoBI methylation fully overlapping the recognition site blocks cleavage; Dcm, CpG and EcoKI methylation have no effect.
Recognition Site 5'-T↓GATCA-3'
3'-ACTAG↑T-5'
Heat Inactivation 80°C for 20 min
Methylation Sensitivity Dam, EcoBI: complete overlap, cleavage blocked; Dcm, CpG, EcoKI: no effect
