Product Name Fast Restriction Enzyme AfeI (Recognition Site: AGC/GCT)
Catalog No. IVDRM-0014
Description Fast Restriction Enzyme AfeI from Alta DiagnoTech is a recombinant rapid restriction endonuclease that achieves accurate DNA cleavage within 5-15 minutes. It is designed for fast digestion of plasmid DNA, PCR products and genomic DNA. A single universal buffer is shared by all enzymes in the fast digestion series to simplify reaction setup, and its excellent activity redundancy allows effortless digestion of excess substrate or difficult templates.
Intended Use Rapid digestion of plasmid DNA, PCR products and genomic DNA for molecular cloning and related molecular biology workflows.
Principle / Technology Recombinant fast restriction endonuclease produced by genetic engineering; recognizes the specific double-stranded DNA sequence 5'-AGC/GCT-3' and cleaves rapidly at 37°C in a universal fast digestion buffer.
Sample Type Plasmid DNA, PCR products, genomic DNA
Performance Range / Specifications Complete digestion within 5-15 min
Supplied at 10 U/μL
Good activity redundancy - readily digests excess substrate or difficult templates
Specificity Specifically recognizes 5'-AGC/GCT-3'; CpG methylation blocks cleavage.
Reaction Conditions / Protocol 1× universal fast digestion buffer; incubate at 37°C
Storage Conditions -25°C to -15°C
Shelf Life 2 years
Package Specifications 20 T
Concentration 10 U/μL
Source / Origin Recombinant (produced by genetic engineering technology)
Expiration Date / Stability Stable for 2 years when stored at -25°C to -15°C.
Compatibility Isoschizomers: Eco47III
Recommended Buffer System A single universal fast digestion buffer is shared across the entire fast restriction enzyme series, simplifying single- and multi-enzyme digestion setups.
Application Notes / Precautions CpG methylation of the recognition site blocks cleavage; verify the methylation status of the substrate DNA when digesting CpG-methylated templates.
Recognition Site 5'-AGC↓GCT-3'
3'-TCG↑CGA-5'
Heat Inactivation 80°C for 20 min
Methylation Sensitivity CpG: cleavage blocked
