Product Name Fast PNGase F, Non-Reducing Format (Rapid N-Deglycosylation, 20 T / 50 T)
Catalog No. IVDRM-0294
Description Conventional peptide-N-glycosidase takes hours to release antibody glycans, and because some species are released faster than others, a partial reaction distorts the apparent glycan profile. That error matters when glycan distribution is a quality attribute of a therapeutic antibody. This preparation is engineered for speed, finishing the digestion in minutes, and it is formulated without reducing agents so the disulphide pattern and higher-order structure survive. Keeping the protein intact is what makes it useful for analysis of the whole molecule by mass spectrometry, and for high-throughput proteomics where a short, uniform step is worth more than absolute completeness at longer incubation.
Intended Use Rapid removal of N-linked glycans from antibodies, immunoglobulins, fusion proteins and other glycoproteins ahead of intact protein mass spectrometry and high-throughput proteomic characterisation.
Principle / Technology The amidase cleaves the amide bond between the asparagine-linked innermost sugar of the glycan and the peptide backbone; without reducing agent and with short incubation the protein disulphide framework stays linked together.
Detection Method Released glycans and deglycosylated protein are analysed by mass spectrometry or chromatography rather than by a signal from the enzyme.
Sample Type Purified antibodies, fusion proteins and glycoproteins in solution.
Performance Range / Specifications Deglycosylation completed within minutes instead of hours; non-reducing conditions preserve disulphide bonds; retains higher-order structure; compatible with intact-protein mass spectrometry workflows.
Specificity Supplied with a 5x concentrated reaction buffer; 20-test vial contains 20 uL of enzyme and 80 uL of buffer; 50-test vial contains 50 uL and 200 uL respectively; denaturation at 75 °C for 5 minutes followed by digestion at 50 °C for 10 minutes.
Reaction Conditions / Protocol Denature the sample for 5 minutes at 75 °C with the supplied buffer, cool, add the enzyme and incubate for about 10 minutes at 50 °C, then move the digest directly into mass spectrometry or chromatographic analysis.
Components / Formulation Enzyme solution and 5x reaction buffer in matched volumes.
Storage Conditions Store at 2 to 8 °C; never freeze.
Shelf Life 1 year from receipt when refrigerated.
Package Specifications 20-test and 50-test presentations, each with enzyme plus 5x buffer.
Product Form Clear enzyme solution with separate concentrated buffer.
Quality Control Each lot is checked for digestion completeness against a reference antibody substrate.
Key Features - Completes deglycosylation in minutes rather than hours
- Non-reducing formulation keeps disulphide bonds intact
- Higher-order structure is preserved for whole-molecule mass analysis
- Supplied with matching 5x reaction buffer
- Suited to high-throughput proteomics workflows
Purity Recombinant enzyme purified to remove contaminating glycosidase activity.
Concentration Supplied ready to use with the companion buffer; see the test count for reaction numbers.
Activity / Unit Definition Defined by complete glycan release from a reference glycoprotein within the stated short incubation rather than by standard units.
Molecular Weight Not separately stated on the source page for this format.
Source / Origin Recombinantly expressed in yeast.
pH Range / Optimal pH Reaction buffer is 20 mM Tris-HCl with 50 mM sodium chloride and 5 mM EDTA at pH 7.5.
Shipping Conditions Shipped cold, kept chilled rather than frozen.
Expiration Date / Stability Stability depends on continuous storage at the stated temperature with the container closed.
Regulatory / Compliance For research use only.
Compatibility Digests feed directly into chromatographic or mass spectrometric analysis without a desalting requirement.
Recommended Buffer System 20 mM Tris-HCl, 50 mM sodium chloride, 5 mM EDTA at pH 7.5 supplied as a 5x concentrate.
Application Notes / Precautions Do not freeze the enzyme; keep it refrigerated; work under the non-reducing protocol so the quaternary structure of the substrate remains intact.
Batch-to-Batch Consistency Fixed production controls and release testing keep performance uniform from lot to lot.
Model Type Rapid non-reducing N-deglycosylation reagent
Starting Material / Tissue Site Yeast-expressed recombinant amidase
Delivery Standard Enzyme vial plus buffer vial
Readouts / Downstream Assays Mass shift of the intact protein after glycan release and the glycan profile obtained downstream.

For in vitro diagnostic use.
