| Product Name | Fast PNGase F for Rapid N-Deglycosylation (20 T / 50 T) |
| Catalog No. | IVDRM-0300 |
| Description | Monitoring glycosylation during antibody manufacture means judging glycan distribution quickly and accurately. Two problems stand in the way with conventional enzyme: digestion takes hours, and glycans are not released at equal rates, so a partial reaction gives a distorted profile that does not represent the real composition. This preparation was developed against exactly that pair of issues. It removes all N-linked glycans within minutes and does so without preference between species, so short incubations still return a truthful distribution, and the result can go straight into chromatography or mass spectrometry. For laboratories running this test repeatedly, the benefit is a shorter workflow that keeps both sensitivity and reproducibility. |
| Intended Use | Rapid and unbiased release of N-linked glycans from antibodies, immunoglobulins, fusion proteins and other glycoproteins during glycosylation analysis. |
| Principle / Technology | As an amide hydrolase it cleaves the bond attaching the glycan to the asparagine side chain, releasing the intact N-linked chain quickly and uniformly. |
| Detection Method | N-linked glycan profile determined by downstream chromatography or mass spectrometry. |
| Sample Type | Purified antibodies, immunoglobulin preparations and glycoproteins in solution. |
| Performance Range / Specifications | Complete deglycosylation within minutes; no preference between glycan species; compatible with immediate downstream chromatography or mass spectrometry; supplied with a 5x buffer. |
| Specificity | Supplied with Fast PNGase F buffer at 5x concentration; 20-test vial holds 20 uL of enzyme and 80 uL of buffer; 50-test vial holds 50 uL and 200 uL respectively. |
| Reaction Conditions / Protocol | Mix the glycoprotein with the 5x buffer, add the enzyme, incubate briefly according to the recommended short schedule, then transfer directly to the analytical step. |
| Components / Formulation | Enzyme solution and 5x reaction buffer. |
| Storage Conditions | Store at 2 to 8 °C; do not freeze. |
| Shelf Life | 1 year from receipt when refrigerated. |
| Package Specifications | 20-test and 50-test presentations with matched buffer volumes. |
| Product Form | Clear enzyme solution with separate concentrated buffer. |
| Quality Control | Each lot is verified for rapid and complete glycan release against a reference antibody. |
| Key Features | - Removes all N-linked glycans within minutes - Unbiased release so the profile reflects true composition - Feeds straight into chromatographic or mass spectrometric analysis - Shortens the workflow without losing sensitivity - Includes the 5x reaction buffer in matched volume |
| Purity | Recombinant amidase free of contaminating protease activity. |
| Concentration | Supplied ready to use with the matching buffer. |
| Activity / Unit Definition | Defined by the ability to release all N-linked glycans within the stated minutes rather than by conventional unit values. |
| Molecular Weight | Not restated on the source page for this format. |
| Source / Origin | Recombinantly expressed in yeast. |
| pH Range / Optimal pH | Buffer composition is 20 mM Tris-HCl at pH 7.5 with 50 mM sodium chloride and 5 mM EDTA. |
| Shipping Conditions | Shipped cold, kept chilled. |
| Expiration Date / Stability | Stability depends on continuous storage at the stated temperature with the container closed. |
| Regulatory / Compliance | Restricted to research applications; not for clinical use. |
| Compatibility | Digests can be loaded directly onto chromatographic or mass spectrometric systems without a clean-up step. |
| Recommended Buffer System | 20 mM Tris-HCl at pH 7.5 with 50 mM sodium chloride and 5 mM EDTA, supplied as a 5x concentrate. |
| Application Notes / Precautions | Do not freeze; keep refrigerated; rely on the validated short incubation rather than extending it, since the bias-free profile is the reason for the shortened protocol. |
| Batch-to-Batch Consistency | Fixed production controls and release testing keep performance uniform from lot to lot. |
| Model Type | Rapid N-deglycosylation reagent |
| Starting Material / Tissue Site | Yeast-expressed recombinant amidase |
| Delivery Standard | Enzyme vial plus 5x buffer vial |
| Readouts / Downstream Assays | Glycan distribution obtained by chromatography or mass spectrometry and completeness of the mass shift. |
