Product Name Endonuclease IV (Nfo) (10 U/uL)
Catalog No. IVDRM-0492
Description Also called Nfo, Endonuclease IV recognises apurinic and apyrimidinic sites in DNA and cuts the phosphodiester bond on the 5 prime side of the AP site to produce a 3 prime hydroxyl group and a 5 prime deoxyribose phosphate end. The enzyme also carries 3 prime diesterase activity that releases phosphoglycolaldehyde, intact deoxyribose 5 prime phosphate and inorganic phosphate from the 3 prime end of DNA. It hydrolyses AP sites to introduce nicks that seed DNA repair in FFPE sample restoration, supports DNA damage detection in single cell gel electrophoresis and alkaline elution or alkaline unwinding, and serves as a single enzyme reagent for RPA isothermal amplification. Supplied at 10 U/uL, it carries no non specific DNase residue and host E. coli genomic DNA at less than 0.05 copies per 10 U.
Intended Use FFPE sample repair, comet assays, alkaline elution and RPA isothermal amplification reagent
Principle / Technology Cleavage of the 5 prime phosphodiester bond at AP sites to give 3 prime hydroxyl and 5 prime dRP ends, with additional 3 prime diesterase activity
Sample Type DNA containing apurinic or apyrimidinic sites; FFPE derived DNA
Reaction Conditions / Protocol Incubate with the AP site containing DNA at 37°C; 100 U leaves the lambda DNA-HindIII digest banding pattern unchanged over 16 hours
Storage Conditions -25 to -15°C
Shelf Life 1 year
Package Specifications 1000 U; 5000 U
Key Features Dual AP endonuclease and 3 prime diesterase activity; low host residue; suited to RPA reagent use
Purity Free of non specific DNase residue; host genomic DNA below 0.05 copies per 10 U
Concentration 10 U/uL
Readouts / Downstream Assays Nick formation at AP sites; comet assay damage readout

For in vitro diagnostic use.
