| Product Name | Endoglycosidase S2 with Polyhistidine Tag, 50 U/uL (1000 U / 5 x 1000 U) |
| Catalog No. | IVDRM-0302 |
| Description | Endoglycosidase S2 targets a very specific bond: the one between the two innermost N-acetylglucosamine residues of the N-glycan carried by immunoglobulin G. That selectivity is useful because antibody Fc glycosylation is the attribute most often under scrutiny, and S2 handles high-mannose, hybrid and biantennary complex structures with equal efficiency. This preparation is expressed in Escherichia coli, is more than 95% pure by SDS-PAGE, and is specified free of exoglycosidase contamination and free of any proteolytic activity, so the observed change comes only from the intended cleavage. A polyhistidine tag is included so the enzyme can be taken out of the reaction afterwards. |
| Intended Use | Removal of Fc N-glycans from immunoglobulin G subclasses during antibody glycan analysis and glyco-engineering work. |
| Principle / Technology | The endoglycosidase cleaves the glycosidic bond linking the two core N-acetylglucosamine residues, leaving a single GlcNAc optionally bearing fucose on the protein. |
| Detection Method | Released glycan and trimmed antibody are analysed by chromatography, electrophoresis or mass spectrometry. |
| Sample Type | Purified immunoglobulin G from mouse and other species, and Fc-bearing constructs. |
| Performance Range / Specifications | Highly specific for IgG N-glycans; hydrolyses high-mannose, hybrid and biantennary complex structures efficiently; no detectable exoglycosidase contamination; no proteolytic activity; His-tagged for removal. |
| Specificity | Specific activity 50 U/uL; purity above 95% by SDS-PAGE; supplied as 1000 U or 5 x 1000 U with a 10x reaction buffer. |
| Reaction Conditions / Protocol | Mix the antibody with the 10x buffer, add the enzyme, incubate at 37 °C, then remove the histidine-tagged enzyme on nickel resin or proceed directly to analysis. |
| Components / Formulation | Enzyme solution and 10x reaction buffer. |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 1 year from receipt when stored as recommended. |
| Package Specifications | Supplied as 1000 U or 5 x 1000 U with matching buffer volumes. |
| Product Form | Clear liquid enzyme with separate concentrated buffer. |
| Quality Control | Released above 95% purity by SDS-PAGE with unit definition verified against native mouse IgG and no detectable exoglycosidase or proteolytic activity. |
| Key Features | - Cleaves specifically between the two core GlcNAc residues of IgG glycans - Efficient across high-mannose, hybrid and biantennary complex structures - No detectable exoglycosidase contamination or proteolytic activity - Above 95% pure by SDS-PAGE - Polyhistidine tag allows easy removal after digestion |

| Recommended Buffer System | 20 mM Tris, 100 mM sodium chloride at pH 7.5 supplied with a 10x reaction concentrate. |
| Application Notes / Precautions | Verify that no exoglycosidase-driven trimming is being read as endoglycosidase activity, and remember the product targets Fc-linked glycans rather than unrelated glycosylation. |
| Batch-to-Batch Consistency | Lot-to-lot consistency is maintained through defined manufacturing specifications; every lot ships with its release record. |
