| Product Name | E. coli DNA Ligase, NAD-Dependent (60 U/µL) |
| Catalog No. | IVDRM-0314 |
| Description | E. coli DNA ligase forms a phosphodiester bond between the adjacent 5'-phosphate and 3'-hydroxyl termini of DNA, with NAD as cofactor and an optimum pH of 7.5-8.0 in Tris-HCl buffer. Activity on blunt-ended substrates is negligible; unlike T4 DNA ligase it joins only protruding ends, although blunt-end ligation becomes possible in the presence of PEG and high concentrations of monovalent cations. The enzyme does not join DNA to RNA or RNA to RNA, and it serves methylation library construction, single-strand DNA library preparation and Okayama-Berg cDNA cloning. |
| Intended Use | Ligation in NGS methylation library preparation, second-strand cDNA synthesis, single-strand DNA library preparation and Okayama-Berg cDNA cloning. |
| Detection Method | Adjacent DNA termini with protruding ends |
| Performance Range / Specifications | Activity 60 U/µL; optimum pH 7.5-8.0 in Tris-HCl buffer; blunt-end activity negligible without PEG and monovalent cations |
