Product Name DNase I, Bovine Pancreas, Type IV Powder
Catalog No. IVDRM-0554
Description An endonuclease isolated originally from pancreas, DNase I targets phosphodiester bonds next to pyrimidines and yields polynucleotides carrying 5 prime phosphate and 3 prime hydroxyl ends with an average product of at least a tetranucleotide. It digests single stranded DNA, double stranded DNA and chromatin at a rate influenced by histones, works best at pH 7 to 8 and depends on calcium ions, and with magnesium present it cuts either strand at random sites while manganese drives cutting of both strands at nearly identical positions. Five millimolar calcium protects the enzyme from hydrolysis. This white to light yellow powder from bovine pancreas is Type IV with purity at or above 80% by Biuret and activity of at least 2000 Kunitz Units per mg of protein, defined at 25°C and pH 5.0 as the amount giving an increase of 0.001 in A260 per mL per minute, and it suits clearing DNA from protein samples, introducing nicks for labelled base incorporation in nick translation and removing DNA contamination during extraction.
