Product Name DNase I, Bovine Pancreas, Type IV, Lyophilised with Calcium Chloride
Catalog No. IVDRM-0553
Description An endonuclease found in many cell types and tissues, DNase I cleaves phosphodiester bonds next to pyrimidines to give polynucleotides with 5 prime phosphate and 3 prime hydroxyl ends, with an average digestion product of at least a tetranucleotide. It digests single stranded DNA, double stranded DNA and even chromatin, whose cutting rate is influenced by histones, works best at pH 7 to 8, depends on calcium ions and is stimulated by divalent metals such as magnesium, manganese, cobalt and zinc, with 5 mM calcium protecting the enzyme from autolysis. In the presence of magnesium it cuts either strand randomly, while with manganese it cuts both strands at nearly the same position. This preparation comes from bovine pancreas as a white to light brown lyophilised powder containing calcium chloride, is Type IV with protein at or above 80% by Biuret and an activity of at least 2000 Kunitz Units per mg of protein.
Intended Use DNA clearance from protein samples, nick insertion for labelled base incorporation and nucleic acid extraction workflows
