| Product Name | dCas9 Nuclease, DNA Binding Only (D10A/H840A) |
| Catalog No. | IVDRM-0510 |
| Description | Obtained by introducing D10A and H840A point mutations into Cas9, dCas9 retains the ability to bind its target DNA sequence but has lost cleavage activity. Guided by sgRNA or crRNA-tracrRNA, it supports transcription level regulation studies in vivo and retrieval of specific target sequences in vitro for downstream experimental work. As with active Cas9, the reaction system must remain free of any contaminating nuclease activity to preserve specificity. |
| Intended Use | Transcription level regulation in vivo and specific target sequence retrieval in vitro |
| Principle / Technology | Guide RNA directed binding of target DNA without cleavage due to combined D10A and H840A inactivation |
| Sample Type | Cells or nucleic acid systems supplied with sgRNA or crRNA-tracrRNA |
| Specificity | D10A and H840A mutations abolish cleavage while preserving target DNA binding |
| Reaction Conditions / Protocol | Combine with sgRNA or crRNA-tracrRNA in a nuclease-free system before application to cells or target sequences |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 1 year |
| Package Specifications | 100 pmol |
| Key Features | Binds target DNA without cutting; supports transcriptional regulation and target sequence capture |
| Readouts / Downstream Assays | Binding or transcription regulation readout at the target locus |
For in vitro diagnostic use.
