| Product Name | dCas9 Nuclease, Catalytically Dead SpCas9 (10 mg/mL) |
| Catalog No. | IVDRM-0585 |
| Description | Derived from Streptococcus pyogenes SpCas9, dCas9 Nuclease carries D10A and H840A substitutions that completely inactivate the HNH and RuvC nuclease domains. The protein keeps its sgRNA-guided ability to bind target DNA specifically but has no DNA cleavage activity and creates no double-strand break, acting as a catalytically dead DNA binding platform. Fusion of effector domains such as KRAB, VP64, fluorescent proteins or methyltransferases to dCas9 enables targeted regulation of chosen genes or loci, supporting CRISPR interference, CRISPR activation, epigenetic editing and genome imaging. Purity is at least 95% with negative endonuclease, exonuclease and RNase activity and endotoxin below 10 EU per mg. |
| Intended Use | CRISPR interference, CRISPR activation, epigenetic editing and genome imaging using a DNA binding platform |
| Principle / Technology | D10A and H840A inactivation of both nuclease domains while sgRNA-guided DNA binding is preserved |
| Sample Type | Cells and DNA targets addressed by sgRNA-guided dCas9 effector fusions |
| Specificity | No DNA cleavage activity and no double-strand break; retains specific target DNA binding |
| Reaction Conditions / Protocol | Deliver the dCas9 effector complex by transfection and read out the targeted regulation |
| Storage Conditions | -25 to -15°C |
