Description A purified rat tail collagen type I solution for coating tissue culture surfaces to promote cell attachment, spreading, and proliferation of primary cells and cell lines requiring extracellular matrix substrates.
Intended Use Creation of biomimetic extracellular matrix coatings on tissue culture plastic and glass surfaces to enhance attachment and growth of primary cells, stem cells, and matrix-dependent cell lines.
Principle / Technology Collagen type I self-assembles into a fibrillar network at neutral pH and 37°C, presenting integrin-binding motifs (RGD sequences) that mediate cell attachment through integrin receptors α1β1 and α2β1.
Detection Method Cell attachment, spreading, and morphology assessed by phase-contrast microscopy; cell proliferation quantified by metabolic activity assay or cell counting

Reaction Conditions / Protocol Dilute collagen stock to desired concentration in sterile 0.02 M acetic acid or PBS, add sufficient volume to cover the culture surface (typical 0.5 mL per well of a 6-well plate), incubate at room temperature for 1 hour or 37°C for 30 minutes, aspirate excess solution, rinse with PBS or culture medium, seed cells
