| Product Name | Cas9 D10A Nickase for Single-Strand DNA Nicking (5 mg/mL) |
| Catalog No. | IVDRM-0586 |
| Description | The SpCas9 D10A mutant from Streptococcus pyogenes, Cas9 D10A Nickase retains only one of the two nuclease centres of the wild-type enzyme and instead produces a nick, a functional single-strand break, in the DNA strand targeted by the sgRNA. Because the enzyme no longer creates a double-strand break it is used where a single-strand cut is wanted, for example paired nickase strategies that improve editing precision. Purity is at least 95% with negative endonuclease, exonuclease and RNase activity and endotoxin below 10 EU per mg. |
| Intended Use | Precision genome editing that requires a DNA single-strand nick instead of a double-strand break |
| Principle / Technology | Mutation of one of the two Cas9 nuclease centres so that only a single-strand nick is generated |
| Sample Type | Cells and DNA targets addressed by sgRNA-guided nicking |
| Specificity | Generates a functional single-strand break; endonuclease, exonuclease and RNase activity negative |
| Reaction Conditions / Protocol | Deliver the nickase ribonucleoprotein complex by transfection and read out the editing outcome |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 100 ug; 1 mg |
| Key Features | Nickase format for paired nicking strategies that raise editing specificity |
| Purity | Greater than or equal to 95% |
| Concentration | 5 to 6 mg/mL |
| Source / Origin | Streptococcus pyogenes |
| Readouts / Downstream Assays | Editing outcomes at the target locus after nicking |
For in vitro diagnostic use.
