| Product Name | BsrDI Restriction Endonuclease (10 U/uL) |
| Catalog No. | IVDRM-0529 |
| Description | Derived from Bacillus stearothermophilus D70, BsrDI is a type IIS restriction enzyme that recognises 5 prime GCAATGNN 3 prime and cuts the top strand two nucleotides downstream of the recognition site while cutting the bottom strand directly after it, producing defined sticky ends for Golden Gate directional assembly. The enzyme runs in its dedicated buffer at 37°C and is heat inactivated at 80°C for 20 minutes. EcoBI methylation overlapping the site blocks cleavage, and Bse3DI together with BseMI are recognised isoschizomers. |
| Intended Use | Directional DNA fragment assembly by Golden Gate cloning |
| Principle / Technology | Type IIS asymmetric cleavage of 5 prime GCAATGNN 3 prime producing sticky ends distal to the site |
| Sample Type | Plasmid DNA and assembly modules for Golden Gate workflows |
| Specificity | EcoBI methylation blocks cleavage; isoschizomers Bse3DI and BseMI |
| Reaction Conditions / Protocol | Digest in 1x dedicated buffer at 37°C; heat inactivate at 80°C for 20 minutes |
