| Product Name | BbvCI Restriction Endonuclease (2 U/uL) |
| Catalog No. | IVDRM-0524 |
| Description | Encoded by the BbvCI gene of Bacillus brevis and produced by recombinant expression in E. coli, BbvCI is a type IIA restriction enzyme that exists as a heterodimer of two different catalytic subunits, each with its own independent cleavage position. The enzyme recognises the non-palindromic sequence CCTCAGC and cuts at positions minus five and minus two relative to the site, generating a three base 5 prime sticky overhang. CpG methylation overlapping the site affects cutting. It runs in the shared rapid series buffer at 37°C and is heat inactivated at 80°C for 20 minutes; digestion beyond 3 hours is not recommended to avoid star activity. |
| Intended Use | Molecular cloning of DNA fragments with asymmetric sticky ends |
| Principle / Technology | Heterodimeric type IIA cleavage at the two asymmetric positions of the non-palindromic CCTCAGC site |
| Sample Type | Double stranded DNA containing CCTCAGC sites |
| Specificity | Cuts the non-palindromic CCTCAGC site at minus five and minus two positions; CpG methylation affects cutting |
| Reaction Conditions / Protocol | Digest in 1x rapid restriction buffer at 37°C for up to 3 hours; heat inactivate at 80°C for 20 minutes |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
