| Description | Engineered from Pyrococcus furiosus DNA polymerase, this high-fidelity enzyme combines 5 prime to 3 prime polymerase activity with 3 prime to 5 prime exonuclease activity and reaches a fidelity 83 times that of Taq DNA polymerase and 9 times that of standard Pfu. Two monoclonal antibodies block both activities at room temperature, giving simple hot-start setup that strongly improves specificity and product yield, and an added extension factor supports long-range amplification of fragments up to 13 kb of lambda DNA and 10 kb of human genomic DNA or cDNA. The optimised 2x reaction buffer with magnesium and dNTPs handles complex templates across a GC content of 30% to 60%, amplification runs at a 68°C annealing temperature and 30 sec per kb extension, and products are blunt ended. |
