| Product Name | alpha2-3,6,8,9 Neuraminidase |
| Catalog No. | IVDRM-0307 |
| Description | alpha2-3,6,8,9 Neuraminidase, also known as alpha2-3,6,8,9 neuraminidase or sialidase, is generated by recombinant expression in E. coli from a gene sourced from Arthrobacter ureafaciens. It hydrolyses sialic acids attached through alpha2-3, alpha2-6, alpha2-8 and alpha2-9 linkages on glycoproteins and oligosaccharides. Cleavage of alpha2-3 and alpha2-6 bonds proceeds faster than that of alpha2-8 and alpha2-9 bonds, so extended incubation is advisable when exhaustive desialylation is the goal. |
| Intended Use | Exhaustive removal of terminal sialic acids from glycoproteins and oligosaccharides, covering all four common sialyl linkage types. |
| Principle / Technology | The enzyme cleaves the ketosidic bond between terminal sialic acid and the subterminal sugar, releasing free Neu5Ac from alpha2-3, alpha2-6, alpha2-8 and alpha2-9 linked substrates. |
| Detection Method | Terminal alpha2-3, alpha2-6, alpha2-8 and alpha2-9 linked sialic acids |
| Sample Type | Glycoproteins, glycopeptides and oligosaccharides |
| Performance Range / Specifications | Supplied at 50 U per uL with purity above 90 percent by SDS-PAGE; cleavage rate for alpha2-3 and alpha2-6 bonds exceeds that of alpha2-8 and alpha2-9 bonds. |
| Specificity | Covers all four common sialyl linkage types; kinetic preference for alpha2-3 and alpha2-6 over alpha2-8 and alpha2-9. |
| Reaction Conditions / Protocol | Incubate the glycoprotein or oligosaccharide substrate with the enzyme in the supplied 10x reaction buffer at 37°C; for substrates rich in alpha2-8 or alpha2-9 linkages extend the reaction to reach complete desialylation. |
| Components / Formulation | alpha2-3,6,8,9 Neuraminidase at 50 U per uL; 10x neuraminidase reaction buffer. Volumes scale with the selected pack size. |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 24 months |
| Package Specifications | 2000 U or 10000 U packs including matched reaction buffer |
| Product Form | Liquid enzyme supplied with reaction buffer |
| Quality Control | Purity verified by SDS-PAGE (greater than 90 percent); activity confirmed with an AMC-conjugated sialylated substrate for every lot. |
| Key Features | Single-enzyme coverage of alpha2-3, alpha2-6, alpha2-8 and alpha2-9 sialyl linkages; recombinant production for reproducible lots. |
| Purity | >90% (SDS-PAGE) |
| Concentration | 50 U/uL |
| Activity / Unit Definition | One unit hydrolyses more than 95 percent of terminal alpha-Neu5Ac from 1 nmol of an AMC-labelled sialylated substrate within 5 minutes at 37°C in a 10 uL reaction. |
| Source / Origin | Recombinant expression in E. coli; gene from Arthrobacter ureafaciens |
