| Product Name | AarI Restriction Endonuclease (5 U/uL) |
| Catalog No. | IVDRM-0522 |
| Description | A type IIS restriction enzyme and a complete isoschizomer of PaqCI, AarI recognises 5 prime CACCTGC(N)4 3 prime and cuts downstream of the site to leave a four base 5 prime overhang. Unlike most type IIS enzymes, AarI needs two or more recognition sites on the substrate, and complete digestion requires the activator supplied with the enzyme; digestion can proceed without it but does not go to completion. CpG methylation overlapping the site affects cutting. The enzyme runs in the shared rapid series buffer at 37°C and is heat inactivated at 80°C for 20 minutes. |
| Intended Use | DNA fragmentation, cloning and genomic analysis with type IIS distal cleavage |
| Principle / Technology | Type IIS cleavage four bases away from 5 prime CACCTGC(N)4 3 prime, requiring multiple sites and an activator for completion |
| Sample Type | Plasmid and genomic DNA with CACCTGC sites |
| Specificity | Complete isoschizomer of PaqCI; requires two or more sites; activator supplied with the enzyme is needed for full digestion; CpG methylation affects cutting |
| Reaction Conditions / Protocol | Digest in 1x rapid restriction buffer at 37°C with the supplied activator; heat inactivate at 80°C for 20 minutes |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 100 U |
| Key Features | Four base 5 prime overhang downstream of the site; activator supported complete digestion |
| Concentration | 5 U/uL |
| Readouts / Downstream Assays | Agarose gel profile of digested DNA |
For in vitro diagnostic use.
