Description	Also known as C2c1, AapCas12b is a tracrRNA:crRNA or sgRNA guided DNA endonuclease from the thermophilic bacterium Alicyclobacillus acidophilus. Guided to double stranded DNA targets carrying a TTN PAM it produces a double strand break with sticky ends, and it cuts single stranded DNA targets independently of a PAM. Once bound to its guide and target DNA it additionally activates collateral trans cleavage that shreds arbitrary single stranded DNA in the reaction, the basis of CRISPR based nucleic acid diagnostics. Its optimal cleavage temperature of 60°C makes it more heat tolerant than AacCas12b and a natural partner for LAMP amplification in integrated constant temperature detection systems.
