YO-PRO-1, also known as oxazole yellow (Oxazole yellow), abbreviated as YP1, is a DNA green fluorescent dye that is impermeable to the membranes of normal animal cells but permeable to the membranes of apoptotic cells. It is commonly used for detecting apoptosis. YO-PRO-1 is a non-membrane-permeable, high-affinity DNA-binding cyanine monomer green fluorescent dye. It exhibits minimal fluorescence when unbound to DNA but emits bright green fluorescence upon binding to DNA. During apoptosis, changes in cell membrane permeability allow YO-PRO-1 to enter the cell and bind to DNA, emitting bright green fluorescence. Therefore, this fluorescent dye is commonly used for analyzing and identifying apoptotic cells. It is important to note that YO-PRO-1 can also stain necrotic cells, so it must be used in combination with propidium iodide (PI), a fluorescent dye specific for necrotic cells, to effectively determine apoptosis. PI, or propidium iodide (PI), is a red fluorescent nucleic acid dye that can only stain necrotic cells with lost membrane integrity and binds to nucleic acids to emit bright red fluorescence. Therefore, the combination of YO-PRO-1 and PI allows simultaneous detection of apoptotic and necrotic cells. Apoptotic cells exhibit green fluorescence, while necrotic cells show both red and green fluorescence positivity. Live cells exhibit little or no fluorescence. |
Application |
This product uses a dual-fluorescence method combining the DNA green fluorescent dye YO-PRO-1 (YP1) and the red fluorescent dye propidium iodide (PI) to detect apoptosis and necrosis. It can be applied to most mammalian cells, including adherent cells and suspension cells. |

Notes |
Fluorescent dyes are prone to quenching. Please take care to avoid exposure to light to slow down fluorescence quenching.
The detection buffer should be used in a sterile environment. Otherwise, it may be contaminated by microorganisms, affecting its efficacy and rendering it unusable.
When using the two fluorescent probes for the first time, they can be aliquoted and stored at -20°C in the dark. Avoid repeated freeze-thaw cycles.
YP1 and PI are irritating to the human body. Handle with care and take appropriate precautions to avoid direct contact with the skin or inhalation.
