Product Name Xanthine Oxidase Activity Fluorometric Assay Kit
Catalog No. ETR-HMM-0077
Description A fluorometric assay kit for measuring xanthine oxidase enzyme activity in biological samples. The assay employs a probe that reacts with hydrogen peroxide, a by-product of the xanthine oxidase-catalyzed conversion of hypoxanthine to xanthine and subsequently to uric acid, to generate a fluorescent signal proportional to enzyme activity. The kit includes a specific inhibitor to distinguish xanthine oxidase activity from background and other peroxide-generating systems.
Intended Use Quantitative measurement of xanthine oxidase activity in serum, tissue homogenates, and cell lysates for studying purine metabolism and oxidative stress.
Principle / Technology Enzyme-coupled fluorometric detection of xanthine oxidase-generated hydrogen peroxide
Detection Method Fluorescence (excitation 535 nm, emission 587 nm)
Sample Type Serum, plasma, tissue homogenates, cell lysates, purified enzyme preparations
Performance Range / Specifications Linear detection range: 0.5–50 mU/mL; reaction time: 30 min
Sensitivity / LOD Detection limit: 0.2 mU/mL
Specificity Specific to xanthine oxidase; minimal interference from xanthine dehydrogenase or aldehyde oxidase
Reaction Conditions / Protocol Mix sample with reaction cocktail (hypoxanthine substrate, fluorescent probe, HRP); incubate 30 min at 37 °C protected from light; read fluorescence
Components / Formulation Hypoxanthine substrate (100×), fluorescent probe (200×), HRP enzyme, xanthine oxidase positive control, assay buffer, allopurinol inhibitor control, H₂O₂ standard
Storage Conditions –20 °C for probe and enzyme; 2–8 °C for buffer and substrate
Shelf Life 6 months at recommended temperature
Package Specifications 100 tests
Product Form Liquid reagents
Quality Control Functional validation with purified xanthine oxidase standard; intra-assay CV ≤8%
Key Features Inhibitor control well enables quantification of xanthine oxidase-specific activity separate from background
Purity Reagent grade; substrate purity ≥98% by HPLC where applicable
Concentration As specified per kit; calibrator and substrate concentrations traceable to reference
Activity / Unit Definition One unit defined as the amount of enzyme that catalyzes conversion of 1 µmol substrate per minute under specified conditions
Molecular Weight Varies by target enzyme as documented
Source / Origin Recombinant or purified from natural sources; synthetic chromogenic/fluorogenic substrates
pH Range / Optimal pH pH 6.0–9.0 depending on specific enzyme assay
Shipping Conditions Cold packs for enzyme components; ambient for buffers
Expiration Date / Stability 6–12 months at recommended storage temperature
Regulatory / Compliance Research use; manufactured under ISO 9001 quality system
Compatibility Compatible with standard microplate readers (absorbance, fluorescence, luminescence modes)
Recommended Buffer System Tris or phosphate buffer optimized per enzyme assay
Application Notes / Precautions Include positive control and inhibitor control in each assay; read within linear range; protect light-sensitive substrates
Batch-to-Batch Consistency Enzyme activity and standard curve linearity within ±15% of reference lot
