| Principle / Technology | Acid phosphatase catalyzes the hydrolysis of p-nitrophenyl phosphate to p-nitrophenol at pH 4.8–5.5; the enzymatic reaction is terminated by raising the pH with sodium hydroxide, which simultaneously develops the yellow color of p-nitrophenol. |
