Description A colorimetric assay kit for measuring acid phosphatase activity in biological samples using p-nitrophenyl phosphate as substrate at acidic pH. Acid phosphatase is a key lysosomal enzyme and biomarker for prostate cancer (prostatic acid phosphatase).
Intended Use Quantitative measurement of acid phosphatase activity in serum, plasma, prostate tissue extracts, seminal fluid, and cell lysates.
Principle / Technology Acid phosphatase catalyzes the hydrolysis of p-nitrophenyl phosphate to p-nitrophenol at pH 4.8–5.5; the enzymatic reaction is terminated by raising the pH with sodium hydroxide, which simultaneously develops the yellow color of p-nitrophenol.
Detection Method Endpoint colorimetric measurement at 405–410 nm
