Product Name Trypan Blue Staining Cell Viability Assay Kit
Catalog No. RESK-0156
Description Trypan Blue is an azo, hydrophilic acidic blue dye. It can penetrate the cell membranes of dead and dying cells, staining them blue, while living cells, owing to the integrity of their cell membranes, exclude the dye. Based on this color change, viable cells (which appear transparent and colorless) can be distinguished from dead cells, and this principle is widely used for the routine assessment of cell viability levels. Trypan Blue can also stain collagen and amyloid. The complex formed by Trypan Blue binding to protein emits red fluorescence. In addition, Trypan Blue (at an appropriate concentration) is commonly used to quench autofluorescence or other fluorescent signals at the cell surface. All components of this kit are supplied sterile and can be used directly in cell experiments. Based on a single-use volume of 0.1 mL, the two kit specifications support 100 and 500 tests, respectively. Cell staining can be observed with a common light microscope, and quantitative counting can be performed simultaneously, enabling relatively accurate quantitative analysis of cell viability.
Intended Use Routine assessment and quantitative analysis of cell viability levels by dye exclusion staining and microscopic counting.
Principle / Technology Trypan Blue is an azo, hydrophilic acidic blue dye that penetrates the compromised cell membranes of dead and dying cells, staining them blue, while living cells with intact cell membranes exclude the dye. Viable cells (transparent and colorless) and dead cells (blue) are distinguished by color under a light microscope and counted for quantitative viability analysis.
Detection Method Light microscopy with quantitative counting of stained (dead) and unstained (viable) cells.
Sample Type Cultured cells (cell suspensions).
Performance Range / Specifications Cell staining is observable with a common light microscope, with simultaneous quantitative counting for relatively accurate quantitative analysis of cell viability. Single-use volume: 0.1 mL per test; the kit specifications support 100 and 500 tests, respectively.
Specificity Selectively stains dead and dying cells with compromised cell membranes blue; living cells with intact membranes exclude the dye and remain colorless.
Reaction Conditions / Protocol 1. Mix the cell sample with the Trypan Blue staining reagent (0.1 mL per test).
2. Observe under a light microscope: viable cells are transparent and colorless, dead cells are blue.
3. Count stained and unstained cells to calculate the cell viability percentage.
Components / Formulation All components are supplied sterile and can be used directly in cell experiments.
Storage Conditions Store at room temperature.
Shelf Life 2 years when stored as directed.
