| Product Name | SRB Cell Proliferation and Cytotoxicity Assay Kit |
| Catalog No. | RESK-0154 |
| Description | The Sulforhodamine B (SRB) colorimetric assay is mainly used to detect cell proliferation. SRB is a pink anionic dye that is readily soluble in water and specifically binds to the basic amino acids of cellular constituent proteins under acidic conditions. It produces an absorption peak at 515 nm, and the absorbance is linearly and positively correlated with cell amount, making it suitable for the quantitative detection of cell number. The kit is simple and rapid to operate, with no need to prepare additional reagents. |
| Intended Use | Quantitative detection of cell proliferation and cytotoxicity by colorimetric measurement of cellular protein content in cultured cells. |
| Principle / Technology | SRB is a pink anionic dye that specifically binds to the basic amino acids of cellular proteins under acidic conditions. Bound dye produces an absorption peak at 515 nm, and the absorbance is linearly and positively correlated with cell amount, enabling quantitative detection of cell number. |
| Detection Method | Colorimetric readout: absorbance measured at 515 nm. |
| Sample Type | Cultured cells. |
| Performance Range / Specifications | Simple and rapid operation; no additional reagents need to be prepared. Absorbance at 515 nm is linearly and positively correlated with cell amount. |
| Specificity | Under acidic conditions, SRB specifically binds to the basic amino acids of cellular constituent proteins. |
| Reaction Conditions / Protocol | 1. Fix the cells with the Fixation buffer. 2. Wash with Washing buffer 1. 3. Stain with the Staining buffer (SRB) under acidic conditions. 4. Wash with Washing buffer 2 to remove unbound dye. 5. Dissolve the bound dye with the Dissolving Solution. 6. Measure the absorbance at 515 nm; the signal is linearly proportional to cell amount. |
| Components / Formulation | Fixation buffer: 50 mL Washing buffer 1: 300 mL Staining buffer: 50 mL Washing buffer 2: 500 mL Dissolving Solution: 100 mL |
