Product Name LDH Cytotoxicity Assay Kit
Catalog No. RESK-0157
Description The LDH Cytotoxicity Assay Kit, also known as an LDH Assay Kit or LDH Release Assay Kit, is a colorimetric kit based on a diaphorase-catalyzed INT chromogenic reaction for measuring lactate dehydrogenase (LDH) activity released upon cytotoxicity, or LDH activity in other samples. Damage to the cell membrane structure caused by apoptosis or necrosis leads to the release of cytoplasmic enzymes into the culture medium, including the enzymatically stable lactate dehydrogenase. Quantifying LDH activity released from cells with compromised plasma membranes enables quantitative analysis of cytotoxicity. LDH release is regarded as an important indicator of cell membrane integrity and is widely used in cytotoxicity testing, serving as a safe and effective alternative to the classical radioactive 51Cr-release assay. The kit is ready-to-use and convenient.
Intended Use Quantitative analysis of cytotoxicity by measuring LDH activity released from damaged cells; also suitable for measuring LDH activity in other samples.
Principle / Technology Under the action of lactate dehydrogenase, NAD+ is reduced to NADH. NADH and INT (2-p-iodophenyl-3-nitrophenyl tetrazolium chloride) are then catalyzed by diaphorase (lipoamide dehydrogenase) to generate NAD+ and the strongly chromogenic product formazan, which produces an absorption peak at 490 nm. Absorbance is linearly and positively correlated with LDH activity, enabling colorimetric quantification of LDH activity in samples such as cell culture supernatants and cell lysates.
Detection Method Colorimetric (absorbance measurement at 490 nm; a reference wavelength of 600 nm or above may optionally be used for dual-wavelength detection).
Sample Type Cell culture supernatant, cell lysates, and other samples containing LDH activity.
Performance Range / Specifications Ready-to-use and convenient format.
Reaction Conditions / Protocol Measure absorbance at 490 nm. A reference wavelength of 600 nm or higher can be used for dual-wavelength detection to correct interference from nonspecific absorption or light scattering (optional, not mandatory). Cytotoxicity (cell death rate) of the sample is calculated relative to the control.
Storage Conditions Store at -25 to -15 °C, protected from light.
Shelf Life 1 year
Package Specifications 500 T
Expiration Date / Stability Valid for 1 year when stored at -25 to -15 °C protected from light.
Regulatory / Compliance For research use only.
Application Notes / Precautions Serum contains LDH; it is recommended that the serum concentration in the culture system not exceed 1%, and heat-inactivated serum is preferred.
LDH released into the cell supernatant can be stored overnight in the refrigerator and assayed the next day; however, to reduce experimental error, fresh sample collection followed by immediate measurement is recommended.
The kit calculates the cell death rate of the sample, which is determined relative to the control.

For research use only, not for clinical use.
