Description Ready-to-use RIPA-based homogenization buffer pre-supplemented with broad-spectrum protease and phosphatase inhibitor cocktail. Balanced ionic/non-ionic detergent combination efficiently solubilizes cytoplasmic, membrane, and nuclear proteins from mammalian tissues. Pre-added inhibitors eliminate preparation variability.
Intended Use Total protein extraction from fresh/frozen mammalian tissues for Western blot, IP, ELISA, proteomics; phosphoprotein analysis; fibrous tissue lysis (heart, muscle, skin).
Principle / Technology RIPA: sodium deoxycholate (ionic) solubilizes membrane proteins; NP-40 (non-ionic) extracts cytoplasmic proteins; SDS (low conc) aids nuclear extraction; comprehensive inhibitors block serine/threonine/cysteine/metallo-proteases and phosphatases
Detection Method 500 µL per 20 mg tissue; homogenize on ice (30 sec, 2-3 pulses); incubate 30 min on ice; centrifuge 14,000×g 20 min at 4 °C; collect supernatant
Sample Type Fresh/frozen tissues: liver, kidney, brain, heart, lung, spleen, muscle, adipose, skin, intestine, tumor biopsies; 10⁷ cells per 500 µL
