Product Name Single Cell Suspension Preparation Reagent Kit (Enzymatic Tissue Dissociation)
Catalog No. ATR-SPS-0019
Description A tissue dissociation kit containing a blend of purified collagenase and neutral protease enzymes optimized for gentle dissociation of solid tissues into viable single cell suspensions. The enzyme mixture preserves cell surface epitopes better than trypsin-based dissociation, making it suitable for downstream flow cytometry, single-cell RNA sequencing, and primary cell culture. The kit includes optimized digestion buffer and stop solution.
Intended Use Enzymatic dissociation of solid tissue specimens into viable single cell suspensions for cytometry and single-cell analysis.
Principle / Technology Collagenase and neutral protease-mediated extracellular matrix digestion with epitope preservation
Detection Method Trypan blue viability counting; flow cytometry viability dye staining
Sample Type Fresh solid tissue specimens: tumor biopsies, spleen, lymph node, lung, liver, and other parenchymal organs
Performance Range / Specifications Typical cell yield: 1–5 × 10⁶ viable cells per 100 mg tissue; viability ≥85%
Sensitivity / LOD Effective dissociation of tissues as small as 10 mg (needle biopsy scale)
Specificity Broad tissue compatibility; collagenase blend digests collagens I–V
Reaction Conditions / Protocol Mince tissue to 1–2 mm pieces; incubate in digestion medium 30–60 min at 37 °C with gentle agitation; filter through 70 µm strainer; wash cells; resuspend in appropriate buffer
Components / Formulation Collagenase blend (Type I, II, IV lyophilized), neutral protease (lyophilized), digestion buffer (HBSS with calcium and magnesium), stop solution (FBS-containing medium), 70 µm cell strainers
Storage Conditions Lyophilized enzymes: –20 °C; buffers: 2–8 °C or room temperature
Shelf Life 12 months for enzymes; 18 months for buffers
Package Specifications 10 tissue dissociations (up to 1 g each) per kit
Product Form Lyophilized enzymes; liquid buffers
Quality Control Viability and cell surface marker retention validated on mouse spleen and human tumor tissue
Key Features Collagenase-based dissociation preserves labile surface markers that are lost with trypsin-based protocols
Purity DNase/RNase free; sterile-filtered where applicable
Concentration As specified on product label
Activity / Unit Definition Sample processing efficiency ≥90% recovery per validation protocol
Molecular Weight Not applicable or as specified for enzyme components
Source / Origin Synthetic and recombinant components as specified
pH Range / Optimal pH As stated in product specification
Shipping Conditions Ambient for buffers; cold packs for enzyme components
Expiration Date / Stability 12–18 months at recommended storage temperature
Regulatory / Compliance Research use; manufactured under ISO 9001 or ISO 13485 quality system
Compatibility Compatible with downstream PCR, mass spectrometry, and immunoassay workflows
Recommended Buffer System Use as directed; components optimized for stated application
Application Notes / Precautions Work quickly to prevent sample degradation; use appropriate protease/phosphatase inhibitors if needed
Batch-to-Batch Consistency Protein precipitation efficiency and analyte recovery within ±15% of reference lot
