Product Name Single Cell Suspension Preparation Buffer, EDTA-Based
Catalog No. ATR-SPS-0010
Description A non-enzymatic cell dissociation buffer formulated with EDTA and gentle surfactants for the preparation of viable single-cell suspensions from adherent cell cultures without trypsin-mediated surface protein cleavage.
Intended Use Detaching adherent cultured cells for flow cytometry, cell sorting by FACS, and single-cell RNA sequencing applications requiring preservation of surface antigen expression and cell viability.
Principle / Technology EDTA chelates calcium and magnesium ions required for integrin-mediated cell adhesion to culture surfaces; gentle agitation releases cells without proteolytic cleavage of surface proteins.
Detection Method Cell viability, surface antigen expression by flow cytometry compared to trypsin control
Sample Type Adherent mammalian cell lines: HeLa, HEK293, CHO, primary fibroblasts, mesenchymal stem cells
Performance Range / Specifications Cell viability >95%; surface antigen preservation >90% compared to scraping control
Sensitivity / LOD N/A
Specificity N/A
Reaction Conditions / Protocol Wash cell monolayer with PBS; add buffer to cover cells; incubate 5–15 min at 37°C; gently tap to release cells; add complete medium to stop; centrifuge and resuspend.
Components / Formulation PBS without Ca2+ and Mg2+, EDTA 5 mM, glucose 5 mM, BSA 0.1%
Storage Conditions 2–8°C
Shelf Life 12 months
Package Specifications 100 mL, 500 mL
Product Form Liquid, sterile-filtered
Quality Control Sterility, cell viability testing, surface antigen preservation validation with CD panel
Key Features Non-enzymatic; preserves surface antigens; gentle cell release; sterile; ready to use; validated for flow cytometry
