| Product Name | Second-Generation DNA Library Prep Kit for Dual Short-Read Sequencing Platforms (PCR and PCR-Free) |
| Catalog No. | REKI-0103 |
| Description | A newly engineered library preparation kit built for the two dominant short-read sequencing platforms. The enzymatic composition has been reworked against the previous generation to improve DNA end repair, A-tailing and adapter ligation efficiency, lifting library conversion noticeably. A novel high-fidelity polymerase raises amplification uniformity and fidelity, and a novel ligase mutant accelerates adapter joining. The kit constructs PCR or PCR-free libraries from 100 pg to 1 ug of gDNA, FFPE, cfDNA or ChIP DNA and connects directly to target-capture workflows. Every reagent passes lot-level performance and stability QC before release. |
| Intended Use | Whole-exome sequencing, metagenomic library construction, automated library preparation on liquid-handling workstations, routine plant and animal genome sequencing, FFPE and cfDNA library workflows, ChIP sequencing. |
| Principle / Technology | Fragmented DNA undergoes end repair, A-tailing and adapter ligation with an engineered ligase mutant, followed by high-fidelity PCR enrichment when a PCR library is required. |
| Sample Type | Genomic DNA, FFPE DNA, cfDNA, ChIP DNA |
| Performance Range / Specifications | Input 100 pg to 1 ug; improved end repair and A-tailing efficiency; significantly higher library conversion than the previous generation. |
| Specificity | Supports PCR and PCR-free library outputs; single-tube per-reaction packaging reduces handling errors. |
| Reaction Conditions / Protocol | Fragment or shear DNA, end repair and A-tail, ligate adapters, enrich by high-fidelity PCR if required, then purify. |
| Components / Formulation | End repair and A-tailing reagent, novel ligase mutant, novel high-fidelity polymerase premix, reaction buffers. |
| Storage Conditions | -25 to -15 °C |
| Shelf Life | 1 year |
| Package Specifications | 8 T, 24 T, 96 T, 96 T (automation) |
| Product Form | Liquid reagents, single-reaction aliquots |
| Key Features | Dual-platform compatibility; target-capture compatible; automation-friendly packaging; stringent lot-to-lot QC. |
| Expiration Date / Stability | Prepared single-use library reagents remain stable for 4 weeks at 4 °C and 2 weeks at 25 °C. |
| Compatibility | mainstream high-throughput sequencing platforms; compatible with target capture panels and automated pipetting workstations |
