Description A ten-fold concentrated ammonium chloride-potassium bicarbonate lysis buffer for the selective removal of erythrocytes from whole blood and tissue samples while preserving leukocyte viability.
Intended Use Lysing red blood cells in whole blood samples prior to flow cytometric immunophenotyping, leukocyte DNA extraction, and other applications requiring purified white blood cell populations.
Principle / Technology Ammonium chloride diffuses into RBCs where it is converted to ammonia and HCl by carbonic anhydrase; the pH drop and osmotic imbalance selectively lyse erythrocytes while leukocytes remain intact due to their different membrane transport properties.
