| Product Name | Red Blood Cell Lysis Solution, Ammonium Chloride-Based, 10x |
| Catalog No. | ATR-SPS-0008 |
| Description | A ten-fold concentrated ammonium chloride-potassium bicarbonate lysis buffer for the selective removal of erythrocytes from whole blood and tissue samples while preserving leukocyte viability. |
| Intended Use | Lysing red blood cells in whole blood samples prior to flow cytometric immunophenotyping, leukocyte DNA extraction, and other applications requiring purified white blood cell populations. |
| Principle / Technology | Ammonium chloride diffuses into RBCs where it is converted to ammonia and HCl by carbonic anhydrase; the pH drop and osmotic imbalance selectively lyse erythrocytes while leukocytes remain intact due to their different membrane transport properties. |
| Detection Method | Leukocyte recovery and viability by flow cytometry, residual RBC count |
| Sample Type | EDTA or heparin anticoagulated whole blood, bone marrow, splenocyte preparations |
| Performance Range / Specifications | >99% RBC lysis efficiency; leukocyte recovery >90%; viability >95% |
| Sensitivity / LOD | N/A |
| Specificity | Selective for RBCs; minimal effect on leukocytes, platelets, and nucleated cells |
| Reaction Conditions / Protocol | Dilute 1:10 with deionized water; add 2 mL to 100 µL whole blood; incubate 5–10 min at room temperature; centrifuge 5 min at 300 × g; wash with PBS. |
| Components / Formulation | Ammonium chloride 1.5 M, potassium bicarbonate 100 mM, EDTA disodium 1 mM at 10x concentration |
| Storage Conditions | 2–8°C |
| Shelf Life | 12 months |
| Package Specifications | 100 mL, 500 mL |
| Product Form | Liquid concentrate (10x) |
| Quality Control | RBC lysis efficiency, leukocyte viability, sterility |
| Key Features | Selective RBC lysis; preserves leukocytes; 10x concentrate for economy; validated for flow cytometry; standard in immunophenotyping |
For research use only, not for clinical use.
