Description A cold non-ionic detergent-based lysis buffer for the rapid isolation of intact nuclei from fresh and frozen tissues and cultured cells for epigenomic applications including ATAC-seq, ChIP-seq, and single-nucleus RNA-seq.
Intended Use Isolating pure nuclei from cells and tissues while preserving chromatin structure and nuclear protein-DNA interactions for downstream epigenomic and transcriptomic analyses.
Principle / Technology NP-40 detergent selectively solubilizes cytoplasmic and plasma membranes while leaving nuclear envelope intact; sucrose cushion or differential centrifugation separates nuclei from cytoplasmic debris.

Reaction Conditions / Protocol Homogenize tissue or resuspend cell pellet in ice-cold nuclei isolation buffer; Dounce homogenize 10–15 strokes; filter through 40 µm strainer; centrifuge at 500 × g for 5 min at 4°C; wash once.
