Live Cell Caspase-3 Activity and Mitochondrial Membrane Potential Detection Reagent |
Catalog No. |
CCAT-HMM-0029 |
Description |
Caspase-3, also known as CPP32, Yama, or apoptin, belongs to the CED-3 subfamily of the caspase family and is one of the most researched caspases in mammalian cells. Caspase-3 is a key enzyme in the apoptosis process, capable of directly and specifically cleaving various substrates, including PARP (poly ADP-ribose polymerase), ICAD (inhibitor of caspase-activated deoxyribonuclease), proCaspase-6, 7, and 9, gelsolin, and fodrin, among others. These protein cleavages mediated by caspase-3 are an important component of the molecular mechanisms of apoptosis. Additionally, caspase-3 is involved in nuclear apoptosis processes, such as chromatin condensation and DNA fragmentation. Furthermore, caspase-3 plays a key role in cell blebbing during apoptosis. |
Application |
This product uses the mitochondrial membrane potential-dependent deep red fluorescent probe Mito-Tracker Deep Red 633 combined with the caspase-3 enzyme fluorescent substrate Caspase-3 Substrate to detect mitochondrial membrane potential and caspase-3 enzyme activity in apoptotic cells. |

Notes |
Avoid microbial contamination, as bacterial or fungal contamination can severely affect the detection results.
After staining, testing should be conducted as soon as possible, as prolonged exposure may lead to an increase in the number of apoptotic or necrotic cells.
GreenNuc Caspase-3 Substrate is not very stable and should be avoided from repeated freeze-thaw cycles (generally not exceeding three times).
Fluorescent substances are prone to quenching. When performing fluorescence observations, minimize observation time and ensure light-protected storage during handling and storage.
For your safety and health, please wear a lab coat and disposable gloves during operation. |
