Product Name Lactate Dehydrogenase (LDH) Activity Assay Kit (Colorimetric)
Catalog No. ETR-HMM-0073
Description A colorimetric assay kit for the quantitative measurement of lactate dehydrogenase activity in biological fluids, cell lysates, and tissue homogenates. LDH is a cytoplasmic enzyme released upon cell membrane damage, making it useful for cytotoxicity assessment.
Intended Use Quantitative measurement of LDH activity in serum, plasma, cell culture supernatant, and tissue extracts for cytotoxicity studies and tissue damage assessment.
Principle / Technology LDH catalyzes the conversion of lactate to pyruvate with concomitant reduction of NAD+ to NADH; the generated NADH reduces a tetrazolium salt to a colored formazan product with absorbance at 490–500 nm.
Detection Method Colorimetric detection at 490–500 nm using a microplate reader or spectrophotometer
Sample Type Cell culture supernatant, serum, plasma, tissue homogenates, and cell lysates
Performance Range / Specifications Linear detection range: 0.1–250 mU/mL LDH activity; reaction time: 30 minutes at 37°C
Sensitivity / LOD ≤0.05 mU/mL minimum detectable LDH activity
Specificity Specific for LDH activity; no interference from other dehydrogenases at physiological concentrations when the supplied substrate mixture is used
Reaction Conditions / Protocol Add 50 μL of sample to 50 μL of reaction mixture in a 96-well plate, incubate at 37°C for 30 minutes protected from light, add 50 μL of stop solution, and measure absorbance at 490 nm
Components / Formulation Lactate substrate solution, NAD+ cofactor, tetrazolium salt (WST), diaphorase enzyme, LDH positive control, assay buffer, stop solution, 96-well plate protocol
Storage Conditions -20°C for enzyme components; assay buffer and substrate at 2–8°C; protect from light
Shelf Life 12 months from date of manufacture
Package Specifications 500 tests in 96-well plate format
Product Form Liquid reagent components requiring mixing before use
Quality Control Each lot validated with purified LDH enzyme standard; linearity R² >0.99; intra-assay CV below 5%
Key Features Simple colorimetric readout compatible with standard microplate readers; high sensitivity enables detection of low-level cytotoxicity; water-soluble formazan product eliminates solubilization steps
