Product Name Lactate Assay Kit (Fluorometric, LDH-Coupled)
Catalog No. CMTR-HMM-0071
Description Fluorometric L-lactate quantification kit based on lactate dehydrogenase-catalyzed oxidation of L-lactate to pyruvate with NAD+ reduction to NADH. Enzyme cycling amplification generates a stable fluorescent signal proportional to lactate concentration. Enables sensitive detection for metabolic flux analysis between glycolysis and oxidative phosphorylation pathways.
Intended Use L-lactate quantification for glycolytic rate measurement, cancer cell metabolic profiling (Warburg effect), cell culture monitoring, and hypoxia/ischemia metabolism research.
Principle / Technology LDH oxidizes L-lactate to pyruvate with NAD+ to NADH; NADH drives fluorogenic probe reduction via enzyme cycling (Ex/Em 530/590 nm); stereospecific for L-lactate
Detection Method Fluorometric; Ex/Em 530/590 nm; colorimetric backup at 570 nm
Sample Type Cell culture supernatant, lysates, tissue homogenates, serum, plasma; 2-20 µL per assay
Sensitivity / LOD Detection range 0.1-20 µM in assay; LOD 0.05 µM L-lactate
Specificity Specific for L-lactate; no cross-reactivity with D-lactate or pyruvate; LDH stereospecific
Reaction Conditions / Protocol Add sample and LDH/NAD+/probe reaction mix; incubate 30 min at RT; read fluorescence
Components / Formulation LDH, NAD+, fluorogenic probe, enzyme amplifier, L-lactate standard (1.0 mM), assay buffer
Storage Conditions Store at -20 °C; protect probe from light
Shelf Life 12 months from manufacture date
Package Specifications 100 assays, 500 assays in 96-well format
Product Form Lyophilized enzymes; liquid buffers and probe
Key Features 50× more sensitive than colorimetric NADH endpoint; LDH stereospecificity; minimal pyruvate interference; enzyme cycling extends linear range
