Description A decrease in mitochondrial membrane potential is an early marker of apoptosis, occurring before the outward flipping of phosphatidylserine on the cell membrane and the activation of caspase proteases. When mitochondrial membrane permeability changes, the membrane potential decreases. This change in membrane potential is caused by the formation of Bax dimers and the activation of Bid, Bak, and Bad, which induce the formation of pores in the mitochondrial membrane. When these pro-apoptotic proteins are activated, mitochondria also release cytochrome C into the cytoplasm.
Application This product is used for apoptosis detection and provides CCCP as a positive control for inducing a decrease in mitochondrial membrane potential.

Notes 1. Before use, centrifuge the product briefly to the bottom of the tube, then proceed with subsequent experiments. 2. JC-1 (100× in DMSO) may solidify and adhere to the bottom, walls, or lid of the centrifuge tube at lower temperatures. Incubate at 20–25°C in a water bath for a short time until completely dissolved before use. 3. When preparing the JC-1 staining working solution, the JC-1 (100× in DMSO) provided in the kit must first be thoroughly dissolved and mixed with sterile diH₂O before adding the 10× Assay Buffer. Do not first prepare the 1× Assay Buffer and then add the JC-1 (100× in DMSO), as this will make it difficult for the JC-1 to dissolve completely, severely affecting subsequent detection. 4. When washing with 1× Assay Buffer after JC-1 staining, maintain the 1× Assay Buffer at approximately 4°C for optimal washing efficacy. 5. After JC-1 staining and washing are complete, subsequent testing should be completed within 30 minutes. Store on ice prior to testing. 6. Fluorescent dyes are prone to photobleaching; please avoid exposure to light to slow down photobleaching. 7. For your safety and health, please wear a lab coat and disposable gloves during operation.
