| Product Name | In-Situ CUT&Tag Library Preparation Kit V2 (single target) |
| Catalog No. | REKI-0084 |
| Description | A library preparation kit for single-target CUT&Tag on short-read sequencing platforms, handling 100 to 100,000 input cells. Compared with conventional chromatin immunoprecipitation sequencing and cleavage-based approaches, the workflow is shorter at about seven hours, simpler to operate, tolerant of low starting material, economical with antibody, and gives higher library yields. Cell capture on lectin beads, antibody incubations, transposase steps, spike-in addition, nuclear lysis, genomic DNA recovery on magnetic beads, amplification and size selection are all supported by the two-box module set, with a DNA standard included for between-sample correction. |
| Intended Use | Protein-DNA interaction mapping; histone modification and transcription factor regulation studies; chromatin profiling from limited cell numbers. |
| Principle / Technology | In-situ transposase tagging converts antibody-bound chromatin fragments into adapter-ligated libraries directly on lectin-captured cells. |
| Sample Type | Human, Chinese hamster, rice cells (100-100,000 cells) |
| Performance Range / Specifications | About 7 hours total; 100 to 100,000 cell input; higher library yield with lower antibody consumption than ChIP-based workflows. |
| Specificity | Single-target in-situ profiling with lectin bead capture. |
| Reaction Conditions / Protocol | Capture cells, incubate primary and secondary antibodies, bind and activate transposase, add spike-in, recover DNA on beads, amplify and size select. |
| Components / Formulation | Lectin beads, DNA extraction reagents, transposase module, spike-in standard, amplification reagents in two boxed modules. |
| Storage Conditions | Box I at 2 to 8 °C; Box II at -25 to -15 °C |
| Shelf Life | 1 year |
| Package Specifications | 4 T, 12 T, 48 T |
| Product Form | Boxed reagent modules |
| Key Features | Fast single-day workflow; built-in spike-in calibration; low input and low antibody consumption. |
| Readouts / Downstream Assays | Single-target chromatin interaction libraries |
