| Description | Hexokinase (HK) is present in many organisms, including bacteria, plants, and mammals, and plays a crucial role in glucose metabolism. Hexokinase phosphorylates glucose to form glucose-6-phosphate, which is used in glycolysis. Hexokinase has four subtypes (HK-I, II, III, and IV). HK-I, HK-II, and HK-III have lower Km values, while HK-IV has a Km value up to 100 times higher. |

| Principle | Glucose is converted to 6-phosphoglucose by hexokinase. Then, 6-phosphoglucose is oxidized by 6-phosphoglucose dehydrogenase to form NADH. NADH reduces the colorless probe to a colored product with strong absorbance at 450 nm. |
