Product Name Hexokinase Activity Assay Reagent
Catalog No. CMTR-HMM-0002
Description Hexokinase (HK) is present in many organisms, including bacteria, plants, and mammals, and plays a crucial role in glucose metabolism. Hexokinase phosphorylates glucose to form glucose-6-phosphate, which is used in glycolysis. Hexokinase has four subtypes (HK-I, II, III, and IV). HK-I, HK-II, and HK-III have lower Km values, while HK-IV has a Km value up to 100 times higher.
Application The product employs a simple, sensitive, and rapid procedure capable of detecting hexokinase activity below 0.1 mU/well.
Principle Glucose is converted to 6-phosphoglucose by hexokinase. Then, 6-phosphoglucose is oxidized by 6-phosphoglucose dehydrogenase to form NADH. NADH reduces the colorless probe to a colored product with strong absorbance at 450 nm.
Assay Type Quantitative
Detection Method Colorimetric
Applicable Instruments Microplate reader
Detection Time 1 h
Sample Type Adherent cells, Serum, Suspension cells, Tissue, Tissue extracts
Detection Limit 0.1 mU/well
Procedure 1. Add samples and standards to wells. 2. Add reaction mixture and incubate for 20-60 minutes. 3. Analyze repeatedly during incubation using a microplate reader.
