| Description | Glutathione is a short peptide composed of three amino acids: cysteine, glutamic acid, and glycine, with concentrations in tissues as high as 1 millimole. Glutathione is the primary low-molecular-weight thiol in cells and plays a key role in defending mammalian cells against oxidative and nitrosative stress. Glutathione levels decrease in the early stages of apoptosis. |

| Principle | This assay can be performed in convenient 96-well or 384-well microplates and can be easily automated without the need for separation steps. The signal can be easily read using a fluorescence microplate reader at Ex/Em = 490/520 nm. |

| Procedure | 1. Add the sample (deproteinized) and standard to the wells. 2. For GSH determination, add Thiol Green to the assay buffer; for total glutathione (GSH + GSSG) determination, also add the GSSG probe. 3. Incubate at room temperature for 10–60 minutes. 4. Analyze using a microplate reader 5. The GSSG level can be calculated by subtracting GSH from the total glutathione level. |
