Description Glutathione peroxidase (GPX) is a widely distributed and important peroxidase enzyme in the body. GPX catalyzes the conversion of reduced glutathione (GSH) into oxidized glutathione (GSSG), thereby reducing toxic hydrogen peroxide into non-toxic hydroxyl compounds, thus protecting biological membranes from ROS damage and maintaining normal cellular function. Additionally, it has protective effects on the liver, enhances immune function, antagonizes the harmful effects of metal ions on the body, and increases radiation resistance.
Application Glutathione peroxidase can catalyze the oxidation of GSH by H₂O₂ to form GSSG, GSH reacts with 5,5'-dithiothreitol-bis-(2-nitrobenzoic acid) to form yellow 2-nitro-5-mercaptobenzoic acid, which has a characteristic absorption peak at 412 nm. The activity of glutathione peroxidase can be characterized by the rate of decrease in absorbance.

Notes If the measured absorbance exceeds 1.0, it is recommended to dilute the crude enzyme solution with extraction solution before measurement and adjust the calculations accordingly; measure the absorbance immediately after color development to avoid prolonged detection time affecting color development and causing measurement errors.
