Description A chromogenic assay kit for the detection and quantification of β-galactosidase activity in cell and tissue lysates, widely used as a reporter gene assay, cellular senescence detection, and lysosomal enzyme activity measurement.
Intended Use Detection and quantification of β-galactosidase enzyme activity in transfected cells (reporter gene assays), senescent cells (SA-β-gal), and lysosomal preparations.
Principle / Technology β-Galactosidase hydrolyzes X-Gal (5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside) to produce galactose and 5-bromo-4-chloro-3-hydroxyindole, which oxidizes to an insoluble blue precipitate (5,5'-dibromo-4,4'-dichloro-indigo).
Detection Method Visual observation of blue color development; spectrophotometric quantification at 615 nm of the solubilized indigo product
