| Product Name | β-Galactosidase Enzyme Activity Detection Kit (X-Gal Substrate) |
| Catalog No. | ETR-HMM-0074 |
| Description | A chromogenic assay kit for the detection and quantification of β-galactosidase activity in cell and tissue lysates, widely used as a reporter gene assay, cellular senescence detection, and lysosomal enzyme activity measurement. |
| Intended Use | Detection and quantification of β-galactosidase enzyme activity in transfected cells (reporter gene assays), senescent cells (SA-β-gal), and lysosomal preparations. |
| Principle / Technology | β-Galactosidase hydrolyzes X-Gal (5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside) to produce galactose and 5-bromo-4-chloro-3-hydroxyindole, which oxidizes to an insoluble blue precipitate (5,5'-dibromo-4,4'-dichloro-indigo). |
| Detection Method | Visual observation of blue color development; spectrophotometric quantification at 615 nm of the solubilized indigo product |
| Sample Type | Cell lysates from transfected mammalian cells or tissue extracts expressing β-galactosidase reporter gene |
| Performance Range / Specifications | Detection range: 0.01–100 mU/mL β-galactosidase activity with linear response |
| Sensitivity / LOD | ≤0.005 mU/mL detection limit with extended incubation (up to 24 hours) |
| Specificity | Specific for β-galactosidase; distinguishes from α-galactosidase and other glycosidase activities through substrate specificity |
| Reaction Conditions / Protocol | Add 50 μL of cell lysate to 50 μL of X-Gal substrate solution, incubate at 37°C for 30 minutes to 24 hours depending on expected activity level, blue color development may be observed visually or quantified spectrophotometrically |
| Components / Formulation | X-Gal substrate concentrate (20 mg/mL in DMF), β-galactosidase reaction buffer, β-galactosidase positive control enzyme, stop solution, lysis buffer |
| Storage Conditions | -20°C for X-Gal concentrate (protect from light and moisture); buffer components at 2–8°C or -20°C |
| Shelf Life | 12 months for X-Gal concentrate; 24 months for buffer components |
| Package Specifications | 500 reactions in 96-well plate format |
| Product Form | Liquid concentrates |
| Quality Control | Each lot tested with purified E. coli β-galactosidase standard; substrate purity verified by HPLC (>99%); background hydrolysis measured |
| Key Features | Simple visual readout suitable for cell staining and tissue sections; quantitative option available for microplate-based assays; widely used across multiple application areas including reporter gene assays and senescence studies |
For research use only, not for clinical use.
