Product Name Elastase Activity Fluorometric Assay Kit
Catalog No. ETR-HMM-0081
Description A fluorometric assay kit designed to measure neutrophil elastase activity in biological samples and inhibitor screening applications. The assay uses a peptide substrate conjugated to a fluorophore whose fluorescence is quenched in the intact state by a nearby quencher molecule. Upon elastase-mediated cleavage of the peptide linker, the fluorophore is released from quenching and emits a quantitative fluorescent signal.
Intended Use Measurement of neutrophil elastase activity in biological fluids and cell lysates, with optional inhibitor profiling capability.
Principle / Technology FRET-based peptide substrate cleavage with fluorescence dequenching
Detection Method Fluorescence (excitation 490 nm, emission 520 nm)
Sample Type Bronchoalveolar lavage fluid, sputum, neutrophil lysates, cell culture supernatant
Performance Range / Specifications Linear range: 0.01–2.0 mU per well; reaction kinetics readable in continuous mode
Sensitivity / LOD Detection limit: 0.005 mU per well
Specificity Substrate cleaved by neutrophil elastase, proteinase 3; minimal cleavage by cathepsin G; specificity confirmed with selective elastase inhibitor (elastatinal)
Reaction Conditions / Protocol Add sample to well; add substrate working solution; read fluorescence kinetically at 37 °C for 30–60 min; fluorescence increase rate proportional to activity
Components / Formulation FRET peptide substrate (lyophilized), elastase positive control, elastatinal inhibitor, reaction buffer, fluorophore reference standard
Storage Conditions –20 °C for substrate, enzyme and inhibitor; 2–8 °C for buffer
Shelf Life 6 months at –20 °C
Package Specifications 100 tests
Product Form Lyophilized substrate, liquid reagents
Quality Control Functional validation with purified human neutrophil elastase; inhibitor control reduces signal ≥90%
Key Features FRET format provides low background and high signal-to-noise ratio, suitable for both endpoint and kinetic reading
Purity Reagent grade; substrate purity ≥98% by HPLC where applicable
Concentration As specified per kit; calibrator and substrate concentrations traceable to reference
Activity / Unit Definition One unit defined as the amount of enzyme that catalyzes conversion of 1 µmol substrate per minute under specified conditions
Molecular Weight Varies by target enzyme as documented
Source / Origin Recombinant or purified from natural sources; synthetic chromogenic/fluorogenic substrates
pH Range / Optimal pH pH 6.0–9.0 depending on specific enzyme assay
Shipping Conditions Cold packs for enzyme components; ambient for buffers
Expiration Date / Stability 6–12 months at recommended storage temperature
Regulatory / Compliance Research use; manufactured under ISO 9001 quality system
Compatibility Compatible with standard microplate readers (absorbance, fluorescence, luminescence modes)
Recommended Buffer System Tris or phosphate buffer optimized per enzyme assay
Application Notes / Precautions Include positive control and inhibitor control in each assay; read within linear range; protect light-sensitive substrates
Batch-to-Batch Consistency Enzyme activity and standard curve linearity within ±15% of reference lot
