Description Methanol-free 16% formaldehyde in sealed glass ampules under argon for ChIP and protein-DNA crosslinking applications. Methanol-free formulation eliminates protein precipitation artifacts. Ampule format ensures fresh, unoxidized reagent for every experiment, avoiding formic acid formation that compromises crosslinking efficiency.
Intended Use ChIP/ChIP-seq protein-DNA crosslinking; 3C/4C/Hi-C chromosome conformation capture; cell/tissue fixation for IHC; reversible crosslinking in protein interaction studies; CLIP-seq RNA-protein crosslinking.
Principle / Technology Formaldehyde crosslinks primary amines (Lys) and thiols (Cys) forming reversible methylene bridges; crosslinks reversed by heat (65 °C) with high salt or proteinase K
Detection Method For ChIP: add to medium to 1% final; incubate 5-15 min RT; quench with 125 mM glycine 5 min; wash with cold PBS
Sample Type Adherent and suspension cells; fresh/frozen tissue; 10⁶-10⁸ cells per ChIP
