Description Caspase 6, also known as Mch-2, is sometimes written as caspase-6 or caspase 6 and was first discovered in human Jurkat cells. The precursor of caspase 6 is cleaved by granzyme B to form activated caspase 6 dimers, and activated caspase 6 has been found to induce apoptosis. Caspase 6 can cleave PARP and keratin-18, as well as the key component protein lamin A on the nuclear envelope. Among the caspase family, only caspase 6 can cleave lamin A. The recognition site for lamin A by caspase 6 is VEID. This assay kit utilizes the specificity of caspase 6 for VEID recognition and has been designed with a corresponding caspase 6-specific chromogenic substrate, Ac-VEID-pNA.
Application This product uses spectrophotometry to detect caspase 6 enzyme activity in cell or tissue lysates or purified caspase 6 enzyme activity.

Notes Ac-YVAD-pNA should be avoided from repeated freeze-thaw cycles. Please note to aliquot appropriately. It is recommended to dilute the sample with water at a 1:1 ratio before determining protein concentration using the Bradford method to minimize interference from DTT on protein concentration measurement. pNA (Chinese name: 4-nitroaniline) is toxic to humans. Handle with extreme caution and ensure proper protective measures to avoid direct contact with the skin or inhalation. pNA (10 mM) may solidify and adhere to the bottom, walls, or lid of centrifuge tubes at low temperatures such as 4°C or ice bath conditions. It can be warmed in a water bath at 20–25°C for a short time until completely dissolved before use. For your safety and health, please wear a lab coat and disposable gloves during handling.
