| Description | The substrate consists of a fluorescent DNA dye coupled to the caspase-3/7 DEVD recognition sequence. The substrate is initially non-fluorescent and crosses the cell membrane into the cytoplasm. In apoptotic cells, caspase-3/7 cleaves the substrate, releasing a high-affinity DNA dye that migrates to the cell nucleus, labels DNA, and emits bright green fluorescence. |

| Notes | 1. Cells can be co-stained with Hoechst 33342 dye at a final concentration of 1 μM to produce blue fluorescence staining of the cell nucleus (excitation/emission: 346/460 nm). 2. SuperView 488 staining can be fixed with formaldehyde but is incompatible with methanol fixation. 3. SuperView 488-stained cells fixed with formaldehyde can be treated with 0.1% Triton X-100 before subsequent staining, but this may reduce the brightness of the staining. 4.
