| Description | The substrate consists of a fluorescent DNA dye coupled to the DEVD recognition sequence of Caspase-3. The substrate is initially non-fluorescent and enters the cytoplasm through the cell membrane. In apoptotic cells, Caspase-3 cleaves the substrate, releasing a high-affinity DNA dye that migrates to the cell nucleus, labels the DNA, and emits bright green fluorescence. Therefore, SuperView 488 Caspase-3 Substrate is dual-functional, capable of detecting Caspase-3 activity while visualizing morphological changes in the cell nucleus during apoptosis. SuperView 488 staining is compatible with formalin fixation and subsequent immunofluorescence experiments. |

| Notes | 1. Before use, briefly centrifuge the product to the bottom of the tube, then proceed with subsequent experiments. 2. Cells can be co-stained with Hoechst 33342 dye at a final concentration of 1 μM to produce blue fluorescence staining of the cell nucleus (Ex/Em = 346/460 nm). 3. SuperView 488 staining is compatible with formaldehyde fixation but incompatible with methanol fixation. 4. SuperView 488-stained cells fixed with formaldehyde can be treated with 0.1% Triton X-100 before subsequent staining, but the brightness of the stained cells may be reduced after treatment. 5. All fluorescent dyes are subject to photobleaching; please avoid exposure to light to slow down photobleaching. 6. For your safety and health, please wear a lab coat and disposable gloves during handling. |
