Product Name Angiotensin-Converting Enzyme (ACE) Activity Colorimetric Assay Kit
Catalog No. ETR-HMM-0078
Description A colorimetric assay kit for determining angiotensin-converting enzyme activity in biological fluids using a synthetic peptide substrate. ACE cleaves the substrate to release a chromogenic product that is quantified by absorbance measurement. The assay is configured for serum and plasma specimens as well as purified protein preparations, and includes a selective ACE inhibitor control to confirm signal specificity.
Intended Use Determination of ACE enzymatic activity in serum and plasma for clinical and pharmacological research applications.
Principle / Technology Enzymatic cleavage of a synthetic chromogenic peptide substrate specific to ACE
Detection Method Colorimetric (absorbance at 405 nm)
Sample Type Human serum, EDTA or heparin plasma; avoid hemolyzed samples
Performance Range / Specifications Linear range: 5–150 U/L; incubation: 30 min at 37 °C
Sensitivity / LOD Lower detection limit: 3 U/L
Specificity Substrate is specifically cleaved by ACE; inhibitor control (captopril-based) blocks ≥95% of signal
Reaction Conditions / Protocol Sample volume: 20 µL; add substrate and incubate 30 min at 37 °C; stop reaction and read at 405 nm
Components / Formulation ACE substrate solution, ACE inhibitor solution (captopril), reaction buffer, stop solution, ACE calibrator
Storage Conditions 2–8 °C for all components
Shelf Life 12 months
Package Specifications 100 tests
Product Form Liquid, ready-to-use reagents
Quality Control Calibrator value assigned against NIST-traceable reference material; within-run precision CV ≤6%
Key Features Inhibitor control included to confirm assay specificity within each run
Purity Reagent grade; substrate purity ≥98% by HPLC where applicable
Concentration As specified per kit; calibrator and substrate concentrations traceable to reference
Activity / Unit Definition One unit defined as the amount of enzyme that catalyzes conversion of 1 µmol substrate per minute under specified conditions
Molecular Weight Varies by target enzyme as documented
Source / Origin Recombinant or purified from natural sources; synthetic chromogenic/fluorogenic substrates
pH Range / Optimal pH pH 6.0–9.0 depending on specific enzyme assay
Shipping Conditions Cold packs for enzyme components; ambient for buffers
Expiration Date / Stability 6–12 months at recommended storage temperature
Regulatory / Compliance Research use; manufactured under ISO 9001 quality system
Compatibility Compatible with standard microplate readers (absorbance, fluorescence, luminescence modes)
Recommended Buffer System Tris or phosphate buffer optimized per enzyme assay
Application Notes / Precautions Include positive control and inhibitor control in each assay; read within linear range; protect light-sensitive substrates
Batch-to-Batch Consistency Enzyme activity and standard curve linearity within ±15% of reference lot
